Daclatasvir is demonstrated to disassemble hyperphosphorylated NS5A proteins, interfering with the operation of fresh HCV replication complexes. It can attach to the RNA of the HCV. It is also reported that daclatasvir also blocks both intracellular viral RNA synthesis and virion assembly/secretion in vivo. Using phosphorylated states as a guide, it is demonstrated that it performs two different functions in HCV RNA replication. The trans-acting activity of NS5A that is hyperphosphorylated regulates HCV assembly and infectious particle formation, while the cis-acting activity of basally phosphorylated NS5A mediates the maintenance of the HCV replication complex. A functional replication complex responsible for viral RNA genome amplification on endoplasmic reticulum membranes includes the viral nonstructural phospoprotein NS5A.

Never take it in quantities or frequencies other than those recommended by your doctor. Ask your doctor or pharmacist to explain any instructions on your prescription label that you do not understand, and carefully follow them. It is usually taken with or without food once a day. Missed dose

Never take two doses on the same day. To continue with your regular dosing schedule, skip the missed dose if you don't remember it until the following day. Take the missed dose as soon as you remember it if you remember the missed dose on the day you were supposed to take it.

Side effects

Any of the following symptoms, including fainting or near-fainting, dizziness or lightheadedness, feeling unwell, weakness, exhaustion, shortness of breath, chest pain, confusion, and memory issues, may occur. If you are taking amiodarone along with sofosbuvir and Daclatasvir and experience any of the following symptoms, seek medical attention right away: fainting or near-fainting weakness fatigue shortness of breath chest pain confusion memory issues. You should also contact your doctor right away if you experience weakness, fatigue, nausea, vomiting, yellow eyes or skin, loss of appetite, or light-colored stools. Serious side effects have been reported with daclatasvir including the following: Slow heart rate (bradycardia). These could be indicators of a serious liver condition. Use daclatasvir in combination with sofosbuvir.

For example if you compare genotypes with breeds of dogs; different breeds of dogs have similar features but also significant differences. Of these three HCV genotypes it is genotype 3 that is the most difficult to cure.
